Abstract

Objective: We sought to investigate the expression of tumor necrosis factor alpha–induced protein 8–like 2 (TIPE2) in peripheral blood mononuclear cells (PBMCs) and that of tissue factor (TF) and thrombospondin-1 (TSP-1) in the plasma of asthma patients and to elucidate the correlation between TIPE2, TF, and TSP-1. 

Methods: A total of 54 patients with asthma treated in the respiratory department of our hospital from September 2017 to March 2019 were randomly enrolled into the study group, while 50 healthy individuals who underwent physical examinations at the same time were recruited into the control group. All participants abstained from food overnight for 12 hours and water for four hours. Then, 5 mL of blood was collected from the median cubital vein under fasting status during the morning of the next day. PBMCs were isolated and stored at −80°C after protein lysis. Similarly, the plasma collected from all participants was stored at −80°C for examination. Plasma TF and TSP-1 levels were determined by enzyme-linked immunosorbent assay, while the levels of TIPE2 and TF in PBMCs were determined by western blotting. The correlation between TIPE2 in PBCMs and TF and TSP-1 in plasma was analyzed by Pearson correlation test. 

Results: As compared with in the control group, plasma TF and TSP-1 levels in the study group were significantly higher (P<0.01), while the expression level of TIPE2 in PBCMs was significantly lower (P<0.01) and the expression level of TF in PBCMs was significantly higher (P<0.01). According to the findings of the Pearson correlation test, the expression level of TIPE2 in the PBCMs of asthma patients was significantly and negatively correlated with the levels of TF and TSP-1 in plasma (P<0.05). 

Conclusions: As compared with in healthy people, the expression level of TIPE2 in the PBMCs of asthma patients is significantly decreased, while the expression of TF in PBMCs and the expression levels of TF and TSP-1 in plasma are significantly increased. The expression level of TIPE2 is certainly associated with the levels of TF and TSP.
