Objective: To investigate the role of miRA-214 in regulating platinum resistance in lung cancer. 

Methods: The IC50 values of cisplatin in lung cancer cell A549 and cisplatin-resistant cell A549/DDP were detected by the CCK8 method; The expression of mira-214 in A549 and A549/DDP cells was detected by real-time quantitative PCR, and the effect of miR-214 on the chemotherapy sensitivity of A549/DDP cells was investigated by CCK8 assay combined with flow cytometry after miR-214 knockout in A549/DDP cells. Biological information analysis and luciferase reporter gene detection were used to verify the regulatory effect of miR-214 on p53. Western blot assay was used to detect the effect of miR-241 on p53, Bcl-2 and Bax expression in A549/DDP cells. 

Results: Compared with A549 cells, A549/DDP cells showed strong resistance to DDP, and the expression of miR-214 in A549/DDP cells was increased (P<.01). Knockout of miR-214 in A549/DDP cells could decrease the IC50 of DDP and enhance the apoptosis induced by DDP. MiR-214 could bind to p53 3’UTR region, thus inhibiting the expression of p53 and consequently promoting Bcl-2 and decreasing the expression of apoptosis-related proteins in Bax. 

Conclusion: miR-214 can promote the resistance of lung cancer cells to DDP by inhibiting the expression of p53.
